The Click-&-Go® EdU 594 Flow Cytometry Assay Kit is a superior alternative to the traditional cell proliferation assays that use radioactive nucleosides, i.e., 3H-thymidine or antibody-based detection of the nucleoside analog bromodeoxyuridine (BrdU). EdU (5-ethynyl-2′-deoxyuridine) is a nucleoside analog of thymidine with a terminal alkyne group that is efficiently incorporated into DNA during active DNA synthesis. In the next step, newly synthesized EdU-labeled DNA is fluorescently labeled through a click reaction with the best-in-class red fluorescent Azide 594 (Alexa Fluor® 594 Azide equivalent). Standard flow cytometry methods are used for determining the percentage of S-phase cells in the population. Sufficient amounts of reagents are provided for 50 assays based on the provided protocol.
