The most accurate proliferation-detection methods are based on the incorporation and measurement of nucleoside analogues in newly synthesized DNA. Traditional cell proliferation assays utilizes radioactive nucleosides, i.e., 3H-thymidine or antibody-based detection of the nucleoside analog bromodeoxyuridine (BrdU). Both methods exhibit several limitations, including troublesome handling of radioactive materials and DNA denaturation step by harsh methods (HCl, heat, or enzymes) to expose the BrdU molecules for anti-BrdU antibody staining. Click-&-Go® EdU 555 Cell Proliferation Assay for imaging a superior alternative to traditional time consuming and difficult to perform consistently cell proliferation assays. Click-&-Go® EdU 555 Cell Proliferation Assay takes advantage of EdU (5-ethynyl-2′-deoxyuridine) is a nucleoside analog of thymidine terminal alkyne group that is efficiently incorporated into DNA during active DNA synthesis. In the next step, EdU-labeled newly synthesized DNA is imaged through a click reaction with best-in-class red fluorescent Azide 555.
