Click-&-Go EdU 488 Cell Proliferation Assay for imaging a superior alternative to traditional cell proliferation assays based radioactive nucleosides, i.e., 3H-thymidine or antibody-based detection of the nucleoside analog bromodeoxyuridine (BrdU). Both methods exhibit several limitations, including troublesome handling of radioactive materials and DNA denaturation step by harsh methods (HCl, heat, or enzymes) to expose the BrdU molecules for anti-BrdU antibody staining. Click-&-Go EdU 488 Cell Proliferation Assay takes advantage of EdU (5-ethynyl-2′-deoxyuridine) is a nucleoside analog of thymidine terminal alkyne group that is efficiently incorporated into DNA during active DNA synthesis. In the next step, EdU-labeled newly synthesized DNA is imaged through a click reaction with best-in-class green fluorescent Azide 488 (Alexa Fluor 488® equivalent). Click-&-Go EdU 488 Cell Proliferation Assay includes blue fluorescent Hoechst 33342 dye for performing cell cycle analysis on samples from adherent cells. Sufficient amount of reagents provided for imaging 50 (18×18) coverslips using 500 µL of reaction buffer per test.
