AZDye™ 488 TFP Ester is an amine reactive, green-emitting dye routinely used to label proteins or antibodies through primary amines (Lys), amine-modified oligonucleotides, and other amine-containing biomolecules. The labeling occurs most efficiently at pH 7-9 and forms a stable, covalent amide bond. AZDye™ 488 conjugates are highly photostable dyes with very high fluorescence quantum yields.

AZDye™ 488 is structurally identical to Alexa Fluor® 488. Its absorption/emission spectra is a perfect match to spectra of many other fluorescent dyes based on sulfonated rhodamine 110 core, including DyLight® 488, Alexa Fluor® 488.

DyLight® and Alexa Fluor® are registered trademark of Thermo Fisher Scientific.

AZDye™ 488 is a water-soluble, highly photostable dye with very high fluorescence quantum yield. Its green fluorescence is pH independent from pH 4 to pH 10. The excitation of AZDye™ ideally suited for the 488 nm line. AZDye™ 488 dye conjugated to a variety of antibodies, peptides, proteins, tracers, and amplification substrates often used for generation of stable signal in imaging and flow cytometry.

2,3,5,6-tetrafluorophenyl (TFP) esters is another type of carboxylic acid derivative that react with primary amines forming covalent amide bond. The amine linkage bond is identical to one formed by the reaction between primary amines and NHS esters or sulfo-NHS esters. However, in most cases, TFP ester displays much better stability toward hydrolysis in aqueous media resulting in more efficiency and better reproducible labeling of biopolymers.
